Summary

Chromosome instability (CIN) causes aneuploidy, driving tumor formation. Numerical CIN (nCIN) can paradoxically suppress tumors, necessitating new research tools for therapeutic development.

Chromosome instability (CIN) leads to aneuploidy, a characteristic of human tumors.
CIN contributes to tumorigenesis via structural changes (translocations, amplifications, deletions) and numerical changes (gains/losses of whole chromosomes, nCIN).

This review focuses on the causes and consequences of numerical CIN (nCIN).

Main Results:

nCIN arises from various defects, including spindle assembly checkpoint compromise, cohesion issues, p53 deficiency, and faulty microtubule-kinetochore attachments.
While nCIN is linked to tumorigenesis, recent findings suggest it can also suppress tumor formation.
Understanding these paradoxical effects requires novel reagents and sophisticated mouse models.

Conclusions:

nCIN has a complex and multifaceted role in cancer, capable of both promoting and suppressing tumor development.
Further investigation using advanced models is crucial to fully elucidate nCIN's mechanisms.
This knowledge could lead to novel cancer therapies targeting nCIN.
