A novel bacterium, Pedobacter boryungensis sp. nov. strain BR-9(T), was discovered in Korean soil. This Gram-negative rod represents a new species within the Pedobacter genus, distinct from known relatives.
Area of Science:
- Microbiology
- Bacteriology
- Phylogenetics
Background:
- Soil ecosystems harbor diverse microbial communities.
- The genus Pedobacter comprises Gram-negative bacteria with various ecological roles.
- Accurate taxonomic classification is crucial for understanding microbial diversity and function.
Purpose of the Study:
- To isolate and characterize a novel bacterial strain from Korean soil.
- To determine the phylogenetic and taxonomic position of the novel isolate.
- To propose a new species within the genus Pedobacter.
Main Methods:
- Isolation and cultivation of bacteria from soil samples.
- Phylogenetic analysis using 16S rRNA gene sequencing.
- Determination of chemotaxonomic properties (fatty acids, menaquinone).
- DNA-DNA hybridization for genetic relatedness assessment.
- Phenotypic characterization for differentiation.
Main Results:
- A novel Gram-negative, non-sporulating, non-flagellated rod, strain BR-9(T), was isolated.
- Phylogenetic analysis placed strain BR-9(T) within the genus Pedobacter, closely related to P. insulae and P. koreensis.
- 16S rRNA gene sequence similarity to related species was below 98.2%.
- Chemotaxonomic data (MK-7, iso-C15:0, summed feature 3) and DNA G+C content (38.5 mol%) were determined.
- Low DNA-DNA relatedness (3.4-4.2%) confirmed genetic distinctness from closely related species.
- Distinct phenotypic properties further supported its novelty.
Conclusions:
- Strain BR-9(T) represents a novel species of the genus Pedobacter.
- The proposed name for this new species is Pedobacter boryungensis sp. nov.
- The type strain is BR-9(T) (=KCTC 23344(T) =CCUG 60024(T)).
