  • Densitometric quantification at 475 nm after derivatization with vanillin-sulfuric acid reagent.
  • Main Results:

    • The method accurately quantifies diterpenoids 1β,3α,8β-trihydroxy-pimara-15-ene (A), 6α,11,12,16-tetrahydroxy-7-oxo-abieta-8,11,13-triene (B), and 2α,19-dihydroxy-pimara-7,15-diene (C).
    • Linear working concentration range of 1-10 μg/spot achieved with high correlation coefficients (r² > 0.998).
    • Method validation confirmed peak purity, precision, robustness, LOD, and LOQ according to ICH guidelines.

    Conclusions:

    • A simple, reproducible, and validated densitometric HPTLC method is established for diterpenoid analysis.
    • This method is suitable for the quantitative analysis and standardization of Premna integrifolia root bark extracts.
    • The developed method ensures reliable quality control of P. integrifolia based on its diterpenoid content.
