Summary

Creatine supplementation and exercise training in rats on a cafeteria diet reduced intramyocellular lipid (IMCL) accumulation. Exercise improved insulin sensitivity, while creatine impacted lipid absorption and muscle FAT/CD36 content.

Area of Science:

  • Metabolic research
  • Nutritional science
  • Exercise physiology

Background:

  • Diet-induced obesity impairs insulin sensitivity and lipid metabolism.
  • Intramyocellular lipids (IMCL) and fatty acid translocase (FAT)/CD36 are implicated in metabolic dysfunction.
  • Creatine and exercise are potential interventions for metabolic health.

Purpose of the Study:

  • To investigate the effects of creatine supplementation and exercise training on insulin sensitivity, IMCL, and FAT/CD36 in rats fed a cafeteria diet.
  • To determine if creatine or exercise differentially impacts these metabolic markers.

Main Methods:

  • Rats were fed a cafeteria diet for 12 weeks, with interventions including exercise training and creatine monohydrate supplementation.
  • Metabolic parameters were assessed using intravenous glucose tolerance tests and muscle tissue analysis.
  • Lipid absorption and content were measured post-study.

Main Results:

  • Exercise training (CAF(TR)) and creatine (CAF(CR)) prevented diet-induced increases in IMCL.
  • Both interventions reduced muscle FAT/CD36 protein content.
  • Creatine supplementation increased fecal fat excretion, suggesting inhibited lipid absorption.
  • Exercise, but not creatine, improved insulin action on muscle glucose transport.

Conclusions:

  • Creatine supplementation inhibits IMCL accumulation in diet-induced obesity by reducing gastrointestinal lipid absorption and muscle FAT/CD36.
  • Exercise-induced IMCL reduction is linked to improved insulin action, whereas creatine's benefits are primarily through lipid metabolism modulation.
