DNA-only Transposons

DNA-only transposons are called autonomous transposons since they code for the enzyme transposase that is required for the transposition mechanism. Insertion of transposons can alter gene functions in multiple ways. They can mutate the gene, alter gene expression by introducing a novel promoter or insulator sequence, introduce new splice sites, and change the mRNA transcripts produced, or remodel chromatin structure.
The donor site from where the transposon is excised is either degraded or...

Summary

This study introduces an efficient method for creating transgenic rats using PiggyBac transposon technology. This novel technique significantly improves the success rate of generating genetically modified rats compared to traditional methods.

Area of Science:

  • Genetics
  • Molecular Biology
  • Animal Models

Background:

  • Generating transgenic rats is crucial for biomedical research.
  • Traditional methods like linear DNA microinjection have limited efficiency.

Purpose of the Study:

  • To describe an efficient protocol for producing transgenic rats.
  • To present an alternative to traditional transgenic rat generation methods.

Main Methods:

  • Coinjection of PiggyBac transposon plasmid DNA and transposase mRNA into fertilized rat eggs.
  • Microinjection of pronuclei followed by immediate transfer to pseudopregnant foster females.

Main Results:

  • Achieved transformation frequencies ranging from 33% to 100% (average ~80%).
