Overview Of Cell Separation And Isolation01:20

Overview Of Cell Separation And Isolation

Cell separation was first achieved in 1964 by S. H. Seal, who separated large tumor cells from the smaller blood cells using filtration. Two years later, Pohl and Hawk performed experiments on how cells respond differently to a nonuniform electric field based on the cell type. Such observations were the inception of cell separation methods, which allow isolating a single cell type from a heterogeneous sample.

Summary

Selecting cells with low expression of transfected cDNA is challenging. Fluorescence-activated cell sorting (FACS) can efficiently isolate cells expressing cell surface proteins from negative backgrounds.

Area of Science:

  • Molecular Biology
  • Cell Biology
  • Immunology

Background:

  • Isolating cells with low expression (<1-5%) of transfected cDNA is difficult.
  • Cell surface or integral membrane proteins are potential targets for selection.
  • Existing methods for cell isolation can be tedious and inefficient.

Purpose of the Study:

  • To describe a method for efficient isolation of cells with low expression of transfected cDNA.
  • To highlight the utility of fluorescence-activated cell sorting (FACS) for cell selection.
  • To provide a solution for researchers facing challenges in cell isolation.

Main Methods:

  • Utilizing fluorescence-activated cell sorting (FACS).
  • Targeting cell surface or integral membrane proteins for selection.
  • Selecting transfected cells from a receptor-negative background.

Main Results:

  • FACS enables powerful selection of cells expressing specific proteins.
  • Efficient isolation is achievable even with low transfection expression efficiency.
  • The method allows for the separation of transfected cells from non-transfected cells.

Conclusions:

  • FACS is a highly effective technique for isolating cells with low expression of transfected cDNA.
  • This method significantly improves the efficiency of cell selection for surface protein expression.
  • Researchers can leverage FACS to overcome challenges in isolating specific cell populations.
