The human immunodeficiency virus type-1 (HIV-1) long-terminal repeat (LTR) contains a novel regulatory element, IST, which initiates short transcripts. The function of these IST-initiated transcripts remains unclear.
Area of Science:
- Molecular Biology
- Virology
- Genetics
Background:
- The human immunodeficiency virus type-1 (HIV-1) long-terminal repeat (LTR) promoter region exhibits complex regulatory sequences.
- Numerous cellular and viral transcription factors interact with these sequences to modulate HIV gene expression.
Purpose of the Study:
- To identify and characterize novel regulatory elements within the HIV-1 LTR.
- To investigate the role of the newly discovered Initiator of Short Transcripts (IST) element in HIV-1 gene regulation.
Main Methods:
- Bioinformatic analysis of the HIV-1 LTR sequence.
- Identification of DNA binding sites for transcription factors YY1 and LSF (or CP-2, or LBP-1) within the LTR.
- Analysis of RNA polymerase II transcription initiation and termination.
Main Results:
- A novel regulatory DNA element, IST, located at position (-)5 to (+)26 of the HIV-1 LTR was identified.
- IST directs RNA polymerase II to synthesize short, nonpolyadenylated transcripts (59-61 nt) that terminate at the TAR element.
- The function of these short transcripts remains to be elucidated.
Conclusions:
- The HIV-1 LTR possesses a previously unrecognized regulatory element, IST.
- IST plays a role in initiating short transcripts, suggesting a complex regulatory mechanism in HIV-1 gene expression.
- Further research is needed to determine the functional significance of IST-initiated transcripts in the HIV-1 life cycle.
