Ribosome Profiling02:24

Ribosome Profiling

Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.

DNA Microarrays02:34

DNA Microarrays

Microarrays are high-throughput and relatively inexpensive assays that can be automated to analyze large quantities of data at a time. They are used in genome-wide studies to compare gene or protein expression under two varied conditions, such as healthy and diseased states. Microarrays consist of glass or silica slides on which probe molecules are covalently attached through surface functionalization. Most commonly, the slides are prepared through the chemisorption of silanes to silica...

Area of Science:

  • Molecular Biology
  • Biochemistry
  • Genetics

Background:

  • RNase protection assay (RPA) is a widely adopted technique for RNA analysis.
  • It enables the detection and quantification of RNA levels within cellular and tissue samples.
  • RPA offers significant advantages compared to traditional methods like Northern blot analysis and RT-PCR.

Purpose of the Study:

  • To detail the methodology and advantages of the RNase protection assay (RPA).
  • To highlight RPA's utility in RNA detection and quantitation.
  • To compare RPA with alternative RNA analysis techniques.

Main Methods:

  • Hybridization of a labeled antisense RNA probe with target RNA in solution.
  • Digestion of excess probe using single-strand specific RNase.
  • Separation of protected hybrids via polyacrylamide gel electrophoresis and visualization through autoradiography.

Main Results:

  • RPA demonstrates high sensitivity and specificity in RNA detection.
  • The assay requires minimal sample input and tolerates RNA degradation.
  • RPA allows for rapid, simultaneous, and high-throughput analysis of multiple RNA transcripts.

Conclusions:

  • RNase protection assay (RPA) is a powerful tool for accurate RNA quantitation.
  • Its advantages make it a preferred method for analyzing RNA expression levels.
  • RPA facilitates the use of custom "designer" probe sets for targeted RNA analysis.
