  • Genetics
  • Biosensor Technology

Background:

  • Lateral flow strip biosensors offer simple, low-cost, and portable DNA genotyping.
  • Current limitations include challenges in multiplexing capabilities.
  • Dry-reagent formats minimize complex laboratory procedures.

Purpose of the Study:

  • To enhance the multiplexing capabilities of lateral flow strip DNA biosensors.
  • To enable rapid, instrument-free visual detection of multiple DNA alleles.
  • To demonstrate a novel approach for multiallele genotyping using enhanced lateral flow strips.

Main Methods:

  • Utilizing polymerase chain reaction (PCR) followed by a single primer extension reaction for all target alleles.
  • Incorporating biotin during primer extension for perfect complementarity to target alleles.

  • Employing polystyrene microspheres functionalized with capture probes on multiple test spots for multiallele detection.
  • Main Results:

    • Demonstrated successful multiallele detection through specific hybridization of primer extension products to capture probes.
    • Achieved enhanced detectability by immobilizing microspheres on the membrane, preventing diffusion and improving hybrid exposure.
    • Validated the specificity and performance of the developed biosensor for multiallele genotyping.

    Conclusions:

    • The enhanced lateral flow strip biosensor significantly improves multiplexing capabilities for DNA genotyping.
    • This approach offers a simple, rapid, and instrument-free method for visual detection of multiple DNA alleles.
    • The developed biosensor is suitable for field testing and small laboratory applications requiring efficient multiallele genotyping.
