Labeling DNA Probes03:31

Labeling DNA Probes

DNA probes are fragments of DNA labeled with a reporter tag to enable their detection or purification. The resulting labeled DNA probes can then hybridize to target nucleic acid sequences through complementary base-pairing, and may be used to recover or identify these regions.
Radioisotopes, fluorophores, or small molecule binding partners like biotin or digoxigenin, are the most widely used reporter tags for labeling DNA probes. These labels can be attached to the probe DNA molecule via...

    Summary
    This summary is machine-generated.

Dehydroergosterol (DHE) effectively labels cell membranes for cholesterol studies, unlike some NBD-cholesterol analogs. This fluorescent probe aids in monitoring cholesterol distribution and trafficking within live cells.

    Area of Science:

    • Cell biology
    • Biochemistry
    • Membrane biophysics

    Background:

    • Cholesterol is vital for cellular membranes, particularly plasma membranes and lipid rafts.
    • Mitochondrial inner membranes and endoplasmic reticulum have limited cholesterol content.
    • Dehydroergosterol (DHE) is a fluorescent cholesterol analog used for live-cell membrane labeling.

    Purpose of the Study:

    • To evaluate the utility of fluorescent cholesterol analogs for monitoring cellular cholesterol.
    • To compare the labeling patterns of DHE and NBD-cholesterol analogs with native cholesterol.

    Main Methods:

    • Utilized dehydroergosterol (DHE) as a fluorescent cholesterol analog.
    • Employed NBD-cholesterol analogs for comparative analysis.
    • Observed and analyzed cellular membrane labeling patterns in live cells.

    Main Results:

    • Dehydroergosterol (DHE) effectively labels cellular membranes, reflecting native cholesterol distribution.
    • NBD-cholesterol analogs exhibited different labeling patterns compared to DHE and native cholesterol.
    • DHE proved useful for monitoring cholesterol distribution and trafficking.

    Conclusions:

    • Dehydroergosterol (DHE) is a reliable fluorescent probe for studying cholesterol distribution in live cells.
    • NBD-cholesterol analogs may not accurately represent native cholesterol localization.
    • DHE facilitates research into cellular cholesterol dynamics and membrane function.
