Summary

Sleep disturbances can precede and follow depression, and some antidepressants may worsen insomnia. Targeting serotonin 5-HT2 receptors may offer a dual treatment for both insomnia and depression.

Background:

Sleep problems are closely linked to depression, often appearing before diagnosis and persisting afterward.
Serotonergic (5-HT) transmission plays a complex role in both sleep and wakefulness, involving various receptor subtypes.

Purpose of the Study:

To review the relationship between sleep disorders and depression.
To explore the role of serotonergic transmission in these conditions.
To identify potential therapeutic strategies targeting serotonin receptors.

Main Methods:

Conducted literature searches in PubMed and MedLine up to March 2010.
Selected 34 relevant articles (reviews, clinical trials) focusing on insomnia, depression, sedative antidepressants, and serotonin.
Analyzed the impact of specific antidepressants and serotonin pathways on sleep.

Main Results:

Insomnia can precede and outlast depression, and treating sleep issues may improve comorbid depression.
Certain antidepressants can induce insomnia or somnolence.
Serotonin 5-HT2 receptor stimulation is implicated in antidepressant-induced insomnia and altered sleep patterns.

Conclusions:

Some antidepressants may exacerbate insomnia, hindering recovery from depression.
5-HT2 receptor antagonists show promise for simultaneously treating comorbid insomnia and depression.
