Background:

The formalinized extracellular antigen (fECA) vaccine derived from B16 F10 melanoma cells contains melanoma-associated antigen B700 as its key component.
Understanding the immune response to B700 antigen is crucial for developing effective melanoma therapies.

Purpose of the Study:

To investigate the immunomodulatory effects of prophylactic intrasplenic inoculation of B700 antigen.
To assess the impact of B700 antigen on both humoral and cellular immune responses against melanoma.

Main Methods:

Mice were inoculated with B700 antigen via intrasplenic injection.
Humoral responses were measured by antibody production and their antiproliferative effects on melanoma cells.
Cellular immune functions, including natural killer (NK) cell and lymphokine-activated killer (LAK) cell activities, were evaluated.

T lymphocyte activation was assessed by CD25 surface antigen and IL-2 R alpha mRNA expression.

Main Results:

B700 antigen inoculation induced antibodies with in vitro antiproliferative effects on B16 F10 melanoma cells.
Splenocyte assays revealed increased NK cell and LAK cell activities post-inoculation.
Intrasplenic B700 antigen administration upregulated CD25 expression on T lymphocytes and IL-2 R alpha mRNA levels.

Conclusions:

Prophylactic B700 antigen vaccination elicits both humoral and cellular immune responses.
These immune responses contribute to decreased primary tumor growth and improved survival rates in melanoma-bearing mice.
B700 antigen shows potential as a therapeutic agent in melanoma treatment.
