Purpose of the Study:

  • To investigate the association between melanocortin-1 receptor (MC1R) gene variants and the severity of facial skin photoaging.
  • To determine if MC1R polymorphism influences the degree of photoaging in middle-aged women.

Main Methods:

  • A cohort of 530 French women (ages 44-70) was analyzed.
  • Facial photoaging severity was assessed by a dermatologist using Larnier's global scale.
  • Logistic regression models adjusted for confounders evaluated the impact of MC1R variants on photoaging grades.

Main Results:

  • 35% of participants were wild-type homozygotes, 49% had one MC1R variant, and 15% had two variants.
  • The presence of two major diminished function MC1R variants was identified as a significant risk factor for severe photoaging (aOR=5.61).

Conclusions:

  • Genetic variations in the MC1R gene are crucial determinants of severe facial skin photoaging.
  • MC1R genotype should be considered in understanding individual susceptibility to sun-induced skin aging.
