Mesenchymal stem cells (MSCs) are adult stem cells that can differentiate into most connective tissue cell types, except for hematopoietic cells, depending upon the source of MSCs. Summary

Mesenchymal stem cells (MSCs) from psoriasis patients show altered profiles, producing more angiogenic and inflammatory factors. This suggests early stem cell changes may contribute to psoriasis development and reduced antioxidant capacity.

Background:

  • Psoriasis pathogenesis involves skin cells, but stem cell behavior is unknown.
  • Inducible nitric oxide synthase (iNOS) and vascular endothelial growth factor (VEGF) are implicated in psoriasis.
  • Mesenchymal stem cells (MSCs) are undifferentiated cells found in skin.

Purpose of the Study:

  • Isolate and characterize skin MSCs from psoriasis patients.
  • Compare psoriatic MSCs with those from atopic dermatitis and healthy subjects.
  • Investigate if MSCs exhibit psoriatic characteristics and if psoriasis originates at the stem cell level.

Main Methods:

  • MSCs were isolated from seven psoriasis, seven atopic dermatitis, and seven healthy subjects.
  • Fluorescence-activated cell sorting was used for characterization.
  • VEGF and nitric oxide (NO) levels were measured; iNOS and VEGF expression analyzed; antioxidant capacity tested.

Main Results:

  • Psoriatic perilesional MSCs produced the highest VEGF.
  • MSCs from lesional psoriatic skin showed the greatest increase in NO.
  • MSCs from lesional psoriatic skin had reduced capacity to neutralize peroxynitrite.

Conclusions:

  • The psoriatic microenvironment alters resident MSCs.
  • MSCs may produce angiogenic and proinflammatory factors in psoriasis.
  • Reduced antioxidant capacity in MSCs may contribute to psoriasis lesion development.
