Favism, a hemolytic reaction to fava beans, is linked to glucose-6-phosphate dehydrogenase deficiency and specific acid phosphatase phenotypes. Certain acid phosphatase phenotypes increase susceptibility to fava bean-induced hemolysis.
Area of Science:
- Biochemistry
- Genetics
- Hematology
Background:
- Favism, a hemolytic reaction to fava beans, is associated with glucose-6-phosphate dehydrogenase deficiency (G6PD(-)) and acid phosphatase locus 1 (ACP(1)) phenotypes.
- Individuals with G6PD(-) and favism show a higher frequency of specific ACP(1) alleles (p(a) and p(c)) compared to the general population.
Purpose of the Study:
- To investigate the hypothesis that toxic substances in Vicia faba (fava beans) trigger hemolysis in specific ACP(1) phenotypes.
- To explore the mechanism involving increased phosphorylation, glycolysis, and reduced glutathione production.
Main Methods:
- Analysis of enzymatic activity (f+s) of different ACP(1) phenotypes with and without Vicia faba extract.
- Comparison of enzymatic activity between ACP(1) phenotypes with low (A, CA, C) and high (B, BA, CB) activity groups.
Main Results:
- Significant differences in enzymatic activity were observed between ACP(1) phenotypes after adding Vicia faba extract.
- ACP(1) phenotypes A, CA, and C exhibited extremely low enzymatic activity in the presence of fava bean extract.
- These low activity phenotypes are associated with reduced glutathione levels, leading to erythrocyte lysis.
Conclusions:
- Specific ACP(1) phenotypes (A, CA, C) are more susceptible to oxidative damage from fava bean components.
- The mechanism involves impaired reduced glutathione production due to altered enzymatic activity, resulting in hemolysis.
- This study elucidates a potential genetic and biochemical basis for favism susceptibility in individuals with certain ACP(1) phenotypes.
