Area of Science:

  • Oncology
  • Dermatology
  • Biomarker Research

Background:

  • Current cutaneous melanoma prognosis relies solely on histopathological features.
  • Morphologically similar tumors exhibit variable clinical behavior, necessitating improved prognostic tools.
  • Numerous potential prognostic biomarkers have been investigated for primary melanoma.

Purpose of the Study:

  • To review selected prognostic biomarkers for cutaneous melanoma.
  • To explore factors hindering the clinical translation of melanoma biomarkers.
  • To identify the gap between melanoma biomarker research and clinical application.

Main Methods:

  • Literature review of prognostic biomarkers in cutaneous melanoma.
  • Evaluation of various detection methods including immunohistochemistry, PCR, CGH, and gene expression arrays.
  • Analysis of biomarker development phases and study design considerations.

Main Results:

  • Despite extensive research, no prognostic biomarkers for melanoma have been successfully translated into clinical practice.
  • Several promising biomarkers have been identified but face significant translation barriers.
  • Factors influencing translation include study design, validation, and regulatory pathways.

Conclusions:

  • There is a significant gap between the identification of prognostic melanoma biomarkers and their clinical implementation.
  • Addressing challenges in biomarker development and study design is crucial for future translation.
  • Further research focusing on robust validation and clinical utility is needed to bridge the current gap.
