Area of Science:

  • Biomaterials Science
  • Enzyme Immobilization
  • Biotechnology

Background:

  • Byssus threads from Mytilus viridis are strong biopolymers used for mussel adhesion.
  • These threads present a potential novel biomaterial support for enzyme immobilization.
  • Characterization and application of byssus threads as a support for urease were explored.

Purpose of the Study:

  • To investigate the use of mussel byssus threads as a support for urease immobilization.
  • To evaluate the biochemical properties and stability of immobilized urease.
  • To compare the performance of urease immobilized on byssus threads from natural and laboratory-sourced mussels.

Main Methods:

  • Byssus threads were collected from mussels in natural (N) and laboratory (L) conditions.

  • Surface characterization using Scanning Electron Microscopy (SEM).
  • Chemical modification of byssus threads followed by urease adsorption and activity assays.
  • Main Results:

    • Immobilized urease exhibited enhanced pH stability compared to free enzyme.
    • Enzymes stored at 6 °C retained 50% activity after 30 days, with greater stability in dry conditions.
    • Optimum temperature increased by 25 °C, and the Michaelis-Menten constant (Km) increased to 2.08 mol for immobilized urease.

    Conclusions:

    • Mussel byssus threads are effective supports for urease immobilization, improving enzyme stability and altering kinetic properties.
    • The biomaterial demonstrates potential for biotechnological applications requiring robust enzyme immobilization.
    • Byssus threads from both natural and laboratory-reared mussels provide viable platforms for enzyme support.
