Summary

Researchers developed a novel survivor selection method using Vibrio cholerae hemolysin (HlyA) to identify hybridoma cells producing neutralizing antibodies against HlyA. This technique successfully isolated five antibody-producing clones and characterized the HlyA protein structure.

Area of Science:

  • Microbiology
  • Immunology
  • Molecular Biology

Background:

  • Vibrio cholerae hemolysin (HlyA) possesses leukocidal activity.
  • Hybridoma technology is used to produce monoclonal antibodies.
  • Characterizing bacterial toxins and their interactions with immune cells is crucial.

Purpose of the Study:

  • To develop a new survivor selection protocol for isolating hybridoma cells expressing neutralizing monoclonal antibodies against HlyA.
  • To characterize the structure and proteolytic cleavage of V. cholerae hemolysin.

Main Methods:

  • Utilized the leukocidal activity of HlyA for survivor selection.
  • Generated a hybridoma bank from mice immunized with HlyA.
  • Screened hybridoma clones for anti-HlyA activity using HlyA treatment.

Related Experiment Videos

  • Performed Western immunoblot and amino-terminal sequence analysis on selected clones.
  • Main Results:

    • Successfully isolated five hybridoma clones (X1-X5) with anti-HlyA activity.
    • Identified three distinct HlyA protein forms (83,200, 71,600, and 60,300 Mr) using Western immunoblot.
    • Determined proteolytic cleavage sites within HlyA and calculated actual molecular masses (79.4, 68.6, and 65.3 kDa).

    Conclusions:

    • The developed survivor selection protocol is effective for identifying hybridoma cells producing neutralizing antibodies against leukotoxic agents.
    • Proteolytic cleavage of HlyA generates distinct functional forms.
    • Amino-terminal microsequencing is a powerful tool for analyzing leukotoxic agents and their interactions.
