IP3/DAG Signaling Pathway01:11

IP3/DAG Signaling Pathway

Membrane lipids such as phosphatidylinositol (PI) are precursors for several membrane-bound and soluble second messengers. Specific kinases phosphorylate PI and produce phosphorylated inositol phospholipids. One such inositol phospholipids are the  phosphatidylinositol-4,5 bisphosphate [PI(4,5)P2], present in the inner half of the lipid bilayer. Upon ligand binding, GPCR stimulates Gq proteins to turn on phospholipase Cꞵ. Activated phospholipase Cꞵ cleaves PI(4,5)P2 and produces two-second...
Interactions Between Signaling Pathways01:19

Interactions Between Signaling Pathways

Signaling cascades usually lack linearity. Multiple pathways interact and regulate one another, allowing cells to integrate and respond to diverse environmental stimuli.
Convergence and divergence, and cross-talk between signaling pathways
Two distinct signaling pathways can converge on a single functional unit, which may either be a single protein or a complex of proteins. The response is either functionally distinct or synergistic between the two pathways but different from the response...

Overview of Cell Signaling

Despite the protective membrane that separates a cell from the environment, cells need the ability to detect and respond to environmental changes. Additionally, cells often need to communicate with one another. Unicellular and multicellular organisms use a variety of cell signaling mechanisms to communicate with the environment.
Cells respond to many types of information, often through receptor proteins positioned on the membrane. For example, skin cells respond to and transmit touch...

Hedgehog Signaling Pathway02:33

Hedgehog Signaling Pathway

The Hedgehog gene (Hh) was first discovered due to its control of the growth of disorganized, hair-like bristles phenotype in Drosophila, much like hedgehog spines. Hh plays a crucial role in the development of organs and the maintenance of homeostasis in both invertebrates and vertebrates. However, while Drosophila has only one Hh protein, mammals have multiple functional Hedgehog proteins - Sonic (Shh), Desert (Dhh), and Indian Hedgehog (Ihh). All of these homologous proteins have adapted to...

Area of Science:

  • Cell Biology
  • Systems Biology
  • Computational Biology

Background:

  • Living systems, including cells, process multiple information sources concurrently.
  • Fluctuating environmental conditions and coexisting signals impact cellular functions.
  • Background cellular activity, termed 'chatter,' can influence signal transmission and cellular responses.

Purpose of the Study:

  • To investigate how external chatter modulates information processing in a human cell signaling network.
  • To analyze the effects of chatter on signal transduction in a fibroblast model.
  • To understand the role of chatter in shaping cellular responses to specific signals.

Main Methods:

  • Utilized a Boolean model to simulate a human fibroblast signal transduction network.
  • Analyzed the impact of varying levels of external chatter on network dynamics.
  • Examined how chatter affects information flow and output activity within the network.

Main Results:

  • External chatter significantly shapes the system's response to information-carrying signals.
  • The level of chatter modulates the activity of network outputs in a complex manner.
  • Chatter effectively determines the pathways through which information flows within the cell signaling network.

Conclusions:

  • Cellular signaling networks exhibit versatility in information processing, adapting to varying levels of background noise.
  • Node dynamics and interactions within the network are not random but confer adaptability.
  • Signaling networks can transition between different information-processing scenarios based on external conditions like chatter.
