Sodium caseinate (CasNa) stimulates granulopoiesis and enhances granulocyte function in vivo. This suggests CasNa may activate the innate immune system, promoting granulocyte production and activity.

Area of Science:

  • Immunology
  • Hematology

Background:

  • Sodium caseinate (CasNa) is known to induce granulocyte differentiation and M-CSF production in vitro.
  • The in vivo effects of CasNa on granulocyte proliferation and activation remain largely unexplored.

Purpose of the Study:

  • To investigate the in vivo induction of granulopoiesis by CasNa.
  • To evaluate the activation of granulocytes in mice treated with CasNa.

Main Methods:

  • BALB/c mice were treated with CasNa intraperitoneally.
  • Granulocyte proliferation was assessed via flow cytometry (BrdU incorporation).
  • Granulocyte activation was measured by phagocytic indices and cytokine levels (G-CSF, GM-CSF) using ELISA.

Main Results:

  • CasNa significantly increased bone marrow granulopoiesis (BrdU+/Gr-1+ cells).

  • Generated granulocytes exhibited enhanced phagocytic activity.
  • Serum levels of G-CSF and GM-CSF, and bone marrow G-CSF, were significantly elevated.
  • Conclusions:

    • CasNa administration in vivo promotes granulopoiesis.
    • The induced granulocytes are functionally active, demonstrating increased phagocytosis.
    • CasNa shows potential as an activator of the innate immune system.
