Summary

Artemether-lumefantrine is a safe and effective treatment for Plasmodium falciparum malaria in all age groups. This artemisinin-based combination therapy demonstrates high cure rates and mild side effects, making it a valuable option for malaria treatment.

Background:

Plasmodium falciparum malaria remains a significant global health challenge.
Artemisinin-based combination therapies (ACTs) are frontline treatments for malaria.
Artemether-lumefantrine (AL) is an FDA-approved ACT for P. falciparum malaria.

Purpose of the Study:

To review the pharmacology, pharmacokinetics, safety, and efficacy of artemether-lumefantrine.
To evaluate AL's effectiveness in treating P. falciparum malaria.
To compare AL's efficacy and safety against other antimalarial treatments.

Main Methods:

Systematic review of English-language articles from PubMed (1947-November 2011).
Inclusion of WHO and CDC malaria treatment guidelines.
Search terms included artemether-lumefantrine, Coartem, and malaria.

Main Results:

Artemether-lumefantrine offers dual mechanisms for rapid and sustained parasite clearance.
Polymerase chain reaction (PCR)-corrected cure rates for AL were noninferior to comparators (28-day: 86-100% vs 51-100%; 42-day: 92-99.3% vs 62-100%).
Adverse effects were generally mild to moderate; adherence challenges were noted but clinical cure rates were comparable.

Conclusions:

Artemether-lumefantrine is a safe and effective antimalarial treatment.
It is suitable for both children and adults with P. falciparum malaria.
AL provides a valuable therapeutic option for malaria management.
