Oral controlled-release pellets of olanzapine (OZ) were successfully designed using sodium alginate and glyceryl palmito-stearate. The optimized formulation (F5) demonstrated stable characteristics and sustained drug release for over 24 hours, ideal for once-daily administration.
Area of Science:
- Pharmaceutical Technology
- Drug Delivery Systems
- Materials Science
Background:
- Olanzapine (OZ) is an antipsychotic medication used for schizophrenia.
- Water-insoluble drugs present challenges for oral controlled-release formulations.
- Developing effective delivery systems is crucial for improving therapeutic outcomes.
Purpose of the Study:
- To design and characterize oral controlled-release matrix pellets of olanzapine (OZ).
- To utilize a blend of Sodium Alginate (SA) and Glyceryl Palmito-Stearate (GPS) as matrix polymers.
- To incorporate Microcrystalline Cellulose (MCC) as a spheronizer enhancer and Sodium Lauryl Sulphate (SLS) as a pore-forming agent.
Main Methods:
- Pelletization technique was employed for formulating drug-loaded pellets.
- Characterization included drug content, particle size distribution, SEM, DSC, FTIR, and XRD.
- Stability studies were conducted on the optimized formulation over 90 days under accelerated conditions.
Main Results:
- Drug content ranged from 93.34-98.12% with mean particle size between 1024-1087µm.
- SEM confirmed spherical pellet morphology; DSC and FTIR indicated drug-polymer compatibility.
- Stability studies confirmed the integrity of the pellets; XRD showed crystalline OZ presence.
Conclusions:
- The optimized formulation (F5) exhibited controlled drug release for over 24 hours via Fickian diffusion.
- Formulation F5 demonstrated excellent stability and drug content.
- The designed pellets are suitable for once-daily olanzapine administration.
