Carboxysomes from Nitrobacter bacteria contain circular DNA. This double-stranded deoxyribonucleic acid (DNA) was found to be a 14-micron molecule within the icosahedral particles.

    Area of Science:

    • Microbiology
    • Molecular Biology
    • Biochemistry

    Background:

    • Carboxysomes are proteinaceous microcompartments found in cyanobacteria and some chemoautotrophic bacteria.
    • Nitrobacter species are important in the nitrogen cycle, oxidizing nitrite to nitrate.

    Purpose of the Study:

    • To isolate and characterize the genetic material within carboxysomes of Nitrobacter winogradskyi and Nitrobacter agilis.
    • To determine the physical properties of the DNA found in these bacterial carboxysomes.

    Main Methods:

    • Isolation of carboxysomes from Nitrobacter winogradskyi and Nitrobacter agilis.
    • Buoyant density centrifugation using ethidium bromide and cesium chloride.
    • Electron microscopy for DNA visualization and measurement.

    Related Experiment Videos

    Main Results:

    • Icosahedral carboxysome particles were successfully isolated.
    • Double-stranded deoxyribonucleic acid (DNA) was detected within the isolated carboxysomes.
    • The particle-bound DNA exhibited a buoyant density of rho 25 = 1.701 g/cm3.
    • Electron microscopy confirmed the DNA as a circular molecule approximately 14 microns in length.

    Conclusions:

    • Nitrobacter carboxysomes encapsulate double-stranded circular DNA.
    • This finding suggests a potential role for carboxysomes in protecting or organizing genetic material in these bacteria.
