MicroRNAs

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns (non-coding regions of a gene) or intergenic regions (stretches of DNA present between genes). Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself, forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA...

Area of Science:

  • Molecular Biology
  • Biochemistry
  • Genetics

Background:

  • MicroRNAs (miRNAs) are short non-coding RNAs regulating cellular processes.
  • Differential expression of miRNAs is observed in neoplasia and other diseases.
  • miRNAs show potential as diagnostic and prognostic biomarkers in various body fluids, particularly blood.

Purpose of the Study:

  • To comprehensively review pre-analytical variables affecting miRNA analysis.
  • To highlight variables specific to circulating and tissue miRNA detection.
  • To ensure reliable miRNA profiling for clinical applications.

Main Methods:

  • Systematic literature review of pre-analytical variables in miRNA analysis.
  • Focus on factors influencing detection in general miRNA analysis.
  • Specific examination of variables impacting circulating and tissue miRNA detection.

Main Results:

  • Numerous pre-analytical variables can significantly influence miRNA detection accuracy.
  • Erroneous results due to pre-analytical factors can alter patient diagnosis and treatment.
  • Systematic studies on pre-analytical variables are scarce but critical.

Conclusions:

  • Standardizing pre-analytical procedures is essential for reliable miRNA biomarker utility.
  • Understanding and controlling pre-analytical variables is crucial for clinical translation of miRNA diagnostics.
  • Further systematic research is needed to optimize miRNA detection methods.
