Summary

Entosis, a novel cell death process, involves live epithelial cells engulfing neighbors to form cell-in-cell structures. This study details methods to quantify entosis formation in vitro.

Area of Science:

  • Cell Biology
  • Cell Death Mechanisms
  • Cytoskeletal Dynamics

Background:

  • Entosis is a recently identified form of non-apoptotic cell death.
  • It is characterized by the engulfment of live cells, forming "cell-in-cell" structures.
  • Entosis is triggered by matrix detachment and actomyosin contractility imbalances.

Purpose of the Study:

  • To describe and validate methods for quantifying entosis.
  • To enable the study of entosis in cells cultured in suspension or soft agar.
  • To facilitate research into the mechanisms driving entosis.

Main Methods:

  • Development of quantitative assays for entosis.
  • Utilizing fluorescence imaging for visualizing cell-in-cell structures.
  • Employing time-lapse microscopy to capture dynamic entotic events.

Main Results:

  • Established protocols for measuring entosis formation.
  • Demonstrated the applicability of methods in different culture conditions (suspension, soft agar).
  • Provided a basis for future mechanistic studies of entosis.

Conclusions:

  • Quantifiable methods for studying entosis are now available.
  • These methods will advance the understanding of this unique cell death pathway.
  • Further research can now explore the regulation and biological significance of entosis.
