Rapid methods for testing fecal coliforms and Salmonella in biosolids offer statistically comparable results to U.S. EPA methods. These faster techniques reduce analysis time and costs for non-regulatory research and monitoring.

Background:

  • U.S. Environmental Protection Agency (U.S. EPA) Methods 1680 and 1682 are standard for testing microorganisms in biosolids.
  • Current U.S. EPA methods require 3-5 days for completion, limiting rapid research and process monitoring.

Purpose of the Study:

  • To evaluate a modified Colilert with Quanti-Tray/2000 method against U.S. EPA Method 1680 for fecal coliforms and E. coli.
  • To assess spread plating on xylose-lysine deoxycholate (XLD) with novobiocin against U.S. EPA Method 1682 for Salmonella sp. in biosolids.

Main Methods:

  • Comparison of a modified Colilert/Quanti-Tray/2000 method with U.S. EPA Method 1680.
  • Comparison of XLD spread plating with novobiocin to U.S. EPA Method 1682.
  • Utilized biosolids samples for microbial enumeration.
