Summary

Peripheral blood regulatory T-cells (Tregs) are elevated in patients with peripheral neuroectodermal tumor (PNET) compared to healthy individuals. Treg levels decrease with chemotherapy but increase upon disease progression.

Background:

  • Regulatory T-cells (Tregs) play a crucial role in immune regulation.
  • Previous studies focused on bone marrow Tregs in peripheral neuroectodermal tumor (PNET), but circulating Tregs remain understudied.

Purpose of the Study:

  • To determine baseline levels of circulating Tregs in PNET patients.
  • To correlate Treg levels with patient characteristics.
  • To observe changes in Treg levels during treatment and upon relapse.

Main Methods:

  • Flow-cytometric analysis of Treg cells (CD4+ CD25+ FoxP3+) was performed on blood samples.
  • Samples were collected from de novo PNET patients at diagnosis, post-chemotherapy, and at relapse/progression.
  • Nine healthy controls were included for comparison.

Main Results:

  • PNET patients exhibited significantly higher absolute Treg counts at baseline compared to healthy controls.
  • Patients with fever showed a significantly higher Treg frequency.
  • Circulating Tregs significantly decreased after neoadjuvant chemotherapy and increased at progression.

Conclusions:

  • Peripheral blood Tregs are elevated in PNET patients compared to healthy controls.
  • Chemotherapy leads to a significant reduction in Tregs, while disease progression is associated with an increase.
