DNA replication during differentiation of cortical cells in Allium roots
1Instituto de Biologia Celular (C.S.I.C.), Velázquez, 144, Madrid-6, Spain.
In Allium roots, mature cortical cells show varying DNA levels (2C or 4C). A proposed root tip signal, decreasing with distance, may regulate this DNA replication in developing cells.
Area of Science:
- Plant biology
- Cell biology
- Root development
Background:
- Cortical cells in Allium roots transition from proliferative to mature states.
- A fraction of these cells replicate DNA during this transition.
- Mature cortical cells exhibit distinct DNA contents (2C and 4C).
Purpose of the Study:
- To investigate the distribution of DNA content in mature cortical cells of Allium roots.
- To propose a regulatory mechanism for DNA replication in these cells.
Main Methods:
- Analysis of DNA content in mature cortical cells from the elongation zone of Allium roots.
- Quantification of cells with 2C (G01) and 4C (G02) DNA content.
Main Results:
- Approximately 60% of mature cortical cells possess 2C DNA content (G01).
- Approximately 40% of mature cortical cells exhibit 4C DNA content (G02).
Conclusions:
- A gradient of a replication-regulating signal, decreasing from the root tip, is hypothesized.
- This signal, acting post-mitotically, could explain the observed distribution of nuclear DNA content in mature cortical cells.
