    Background:

    • A previously described monoclonal antibody, 3D6, targets a T cell receptor (Ti) epitope on the HPB-ALL T leukemic cell line.
    • This 3D6 antibody cross-reacts with a small percentage (2-10%) of T cells from healthy individuals.

    Purpose of the Study:

    • To establish and characterize T cell clones that express the 3D6 epitope.
    • To investigate the functional and phenotypic diversity among 3D6-positive T cells.
    • To explore the relationship between different Ti epitopes and T cell characteristics.

    Main Methods:

    • Two protocols were used to establish 3D6+ T cell clones from donor HY.
    • Protocol 1 involved stimulating T cells with JY cells, enriching 3D6+ cells via rosetting, and cloning.
    • Protocol 2 involved stimulating T cells with the 3D6 antibody, expanding them in IL-2, activating with Daudi or TT cells, and cloning.
