Summary

Zoledronic acid (ZOL) significantly reduced circulating endothelial cells (CECs) and circulating endothelial precursor cells (CEPs) in early prostate cancer patients. This suggests ZOL may have antiangiogenic effects by modulating CECs and CEPs.

Background:

  • Zoledronic acid (ZOL) is known to possess antiangiogenic properties.
  • Measuring circulating endothelial cells (CECs) and circulating endothelial precursor cells (CEPs) is a potential method for monitoring antiangiogenic therapies.

Purpose of the Study:

  • To investigate the effect of ZOL on CEC and CEP levels in patients with localized prostate cancer.
  • To explore the potential antiangiogenic mechanism of ZOL in early prostate cancer.

Main Methods:

  • Ten patients with low-risk prostate adenocarcinoma received intravenous ZOL infusions at 0, 28, and 56 days.
  • Blood samples were collected at multiple time points (baseline, 72 hours, 28 days, 56 days, 84 days) for CEC/CEP quantification.
  • Flow cytometry was used to directly quantify CEC and CEP levels.

Main Results:

  • A statistically significant reduction in the mean percentage of CECs was observed at day 3 post-ZOL initiation (p = 0.014).
  • A significant reduction in the mean percentage of CEPs was observed at day 84 (p = 0.012).

Conclusions:

  • Preliminary findings suggest ZOL may exert an antiangiogenic effect in early prostate cancer.
  • The modulation of CEPs and CECs by ZOL supports its potential role in antiangiogenic therapy for prostate cancer.
