Daptomycin shows rapid bactericidal action against Bacillus anthracis by disrupting membrane potential, not integrity. This makes it a potential alternative therapy for anthrax infections.
Area of Science:
- Microbiology
- Infectious Diseases
- Pharmacology
Background:
- Anthrax is a serious infectious disease caused by Bacillus anthracis.
- Novel therapeutic strategies are needed to combat Bacillus anthracis infections.
Purpose of the Study:
- To evaluate daptomycin's efficacy against Bacillus anthracis.
- To elucidate the bactericidal mechanisms of daptomycin against Bacillus anthracis.
Main Methods:
- Determined Minimum Inhibitory Concentration (MIC) values for daptomycin.
- Assessed cell membrane potential using flow cytometry and fluorescent dyes.
- Measured membrane integrity via To-Pro-3 iodide staining and electron microscopy.
- Quantified potassium (K+) efflux and sodium (Na+) influx using fluorescent probes.
Main Results:
- Daptomycin demonstrated rapid bactericidal activity against vegetative Bacillus anthracis (MIC: 0.78 μg/mL).
- Daptomycin inhibited the growth of germinated Bacillus anthracis spores.
- Daptomycin dissipated Bacillus anthracis membrane potential, causing ion flux (K+ efflux, Na+ influx) without compromising membrane integrity.
- Unlike daptomycin, ciprofloxacin and penicillin G did not affect membrane potential; penicillin G disrupted membrane integrity.
Conclusions:
- Daptomycin effectively kills Bacillus anthracis by reducing membrane potential, offering a new therapeutic avenue.
- Daptomycin presents a promising alternative treatment option for anthrax infections.
