    Summary

Alterations in the c-myc oncogene, including structural changes and copy number variations, are implicated in human thyroid cancer development. These c-myc oncogene changes may serve as valuable tumor markers for thyroid neoplasms.

    Area of Science:

    • Oncology
    • Molecular Biology
    • Genetics

    Background:

    • The c-myc oncogene plays a critical role in cell growth and differentiation.
    • Dysregulation of the c-myc oncogene is frequently observed in various cancers.

    Purpose of the Study:

    • To investigate the potential role of the c-myc oncogene in human thyroid neoplastic transformation.
    • To analyze structural, methylation, and copy number alterations of the c-myc oncogene in thyroid tumors.

    Main Methods:

    • Southern blotting technique was employed to analyze DNA structure.
    • Dot blot hybridization was used to assess gene copy number and methylation status.
    • Analysis was performed on DNA extracted from normal thyroid tissue and thyroid carcinomas.

    Related Experiment Videos

    Main Results:

    • Four out of six thyroid carcinomas exhibited abnormal c-myc DNA structures, including mutations and deletions.
    • A 5 kb deletion involving the first exon of c-myc was observed in one carcinoma and its corresponding white blood cells.
    • Three carcinomas showed a double copy number of the c-myc oncogene.
    • Undermethylation of the c-myc oncogene was a consistent finding in all examined carcinomas.

    Conclusions:

    • Alterations in the c-myc oncogene, such as structural abnormalities and copy number changes, are associated with malignant transformation of the human thyroid.
    • These c-myc oncogene alterations may function as potential biomarkers for thyroid cancer detection and diagnosis.
