The ATOH7 gene influences optic disc size, a key glaucoma trait. However, this study found no independent association between ATOH7 and the cup-to-disc ratio (CDR), suggesting its effect is mediated by disc size.
Area of Science:
- Ophthalmology
- Genetics
- Optic Nerve Research
Background:
- The ATOH7 gene is linked to glaucoma and optic disc characteristics.
- Optic disc size and cup-to-disc ratio (CDR) are relevant glaucoma phenotypes.
- The precise relationship between optic disc size and glaucoma requires further elucidation.
Purpose of the Study:
- To determine if ATOH7 is independently associated with CDR.
- To investigate the relationship between ATOH7 and optic disc size.
- To differentiate the effects of ATOH7 on CDR versus optic disc size.
Main Methods:
- Association analysis of the ATOH7 gene region in two distinct populations (Blue Mountains Eye Study and TwinsUK cohort).
- Linear regression models were used to analyze disc size and CDR, adjusting for age, sex, and intraocular pressure (IOP).
- A specific single nucleotide polymorphism (SNP), rs7916697, was examined for its association with disc size and CDR under various adjustment models.
Main Results:
- A significant association was detected between the SNP rs7916697 and optic disc size.
- This SNP was also associated with CDR when adjusted for age, sex, and IOP.
- The association between rs7916697 and CDR was not significant when disc size was included as a covariate.
Conclusions:
- The ATOH7 gene is associated with optic disc size.
- The association of ATOH7 with CDR is likely mediated through its effect on optic disc size.
- ATOH7 does not appear to have an independent effect on CDR beyond its influence on disc size.
