Area of Science:

  • Biochemistry
  • Biomedical Engineering
  • Hematology

Background:

  • Free hemoglobin causes nephrotoxicity and precipitates easily.
  • Developing safe and effective hemoglobin-based oxygen carriers is crucial.

Purpose of the Study:

  • To covalently modify hemoglobin and dextran-hemoglobin.
  • To evaluate the oxygen-releasing properties and safety of modified conjugates.

Main Methods:

  • Reductive alkylation under aerobic conditions.
  • Modification using periodated inositol tetrakisphosphate.
  • Testing in macaques with 90% erythrocyte exchange transfusion.

Main Results:

  • Modified hemoglobin and dextran-hemoglobin showed right-shifted oxygen dissociation curves.
  • The dextran-hemoglobin conjugate ensured adequate oxygen delivery in macaques.
  • The conjugate exhibited no nephrotoxicity and improved stability.

Conclusions:

  • Covalent modification enhances hemoglobin's oxygen-carrying capacity and safety.
  • Dextran-hemoglobin conjugates are promising blood substitutes with reduced adverse effects.
