The novel transcription factor ZNF580 regulates endothelial nitric oxide synthase (eNOS) expression and endothelial cell functions. ZNF580 mediates these effects through the transforming growth factor-β1 (TGF-β1) signaling pathway.
Area of Science:
- Molecular Biology
- Cell Biology
- Biochemistry
Background:
- Transforming growth factor-β1 (TGF-β1) is implicated in regulating endothelial nitric oxide synthase (eNOS) in endothelial cells.
- ZNF580, a C2H2 zinc-finger nuclear transcription factor, is a novel factor with potential roles in TGF-β1 signaling.
- Understanding the interplay between ZNF580, TGF-β1, and eNOS is crucial for vascular biology.
Purpose of the Study:
- To investigate whether ZNF580 mediates eNOS expression.
- To determine ZNF580's role in endothelial cell migration and proliferation.
- To elucidate the involvement of the TGF-β1/Smad2/ZNF580/eNOS signaling pathway.
Main Methods:
- ZNF580 and eNOS expression were modulated (overexpression/downexpression) in EAhy926 cells.
- TGF-β1 treatment was applied in concentration- and time-dependent manners.
- The effect of ALK5 inhibitor SB431542 on TGF-β1-induced changes was assessed.
- Endothelial cell growth and mobility assays were performed.
Main Results:
- ZNF580 overexpression enhanced eNOS expression, while downexpression decreased it.
- TGF-β1 downregulated both ZNF580 and eNOS at mRNA and protein levels.
- The ALK5 inhibitor SB431542 blocked TGF-β1-induced downregulation of ZNF580 and eNOS.
- ZNF580 overexpression counteracted TGF-β1's inhibitory effects on cell growth and migration.
Conclusions:
- ZNF580 mediates eNOS expression in endothelial cells.
- ZNF580 plays a significant role in endothelial cell migration and proliferation.
- The TGF-β1/ALK5/Smad2/ZNF580/eNOS pathway is critical for vascular endothelial cell function.
