    Summary

    This study developed an in vitro method using THP-1 cells to identify chemical sensitizers. Measuring tumor necrosis factor-alpha (TNF-alpha), interleukin-1beta (IL-1beta), and interleukin-8 (IL-8) effectively distinguished allergens from non-allergens.

    Background:

    • Skin sensitization is a significant health concern.
    • Current methods for assessing sensitization have limitations.
    • Developing reliable in vitro alternatives is crucial for chemical safety assessment.

    Purpose of the Study:

    • To establish an in vitro test method for evaluating chemical sensitization.
    • To identify reliable cytokine markers for sensitization using THP-1 cells.
    • To differentiate between chemical allergens and non-allergens.

    Main Methods:

    • THP-1 cells were differentiated into macrophages.
    • Cells were exposed to known allergens (DNCB, NiSO4, PPDA, K2Cr2O7, TDI) and non-allergens (SDS, IPA).
    • Cytokine production (IL-6, TNF-alpha, IL-1beta, IL-8) was measured using ELISA and Cytometric Bead Array.

    Main Results:

    • Allergens significantly increased TNF-alpha, IL-1beta, and IL-8 production.
    • IL-6 levels were also elevated by allergens but notably by non-allergen SDS, suggesting it's not a reliable marker.
    • TNF-alpha, IL-1beta, and IL-8 showed distinct patterns differentiating allergens from non-allergens.

    Conclusions:

    • The THP-1 cell-based assay effectively distinguishes chemical allergens from non-allergens.
    • TNF-alpha, IL-1beta, and IL-8 are proposed as reliable biomarkers for identifying potential sensitizing chemicals.
    • This in vitro method offers a promising alternative for sensitization testing.
