This study synthesized methotrexatum intercalated layered double hydroxides (MTX/LDHs) using reverse microemulsions. The drug efficacy of these MTX/LDHs hybrids was found to be closely associated with the dispersion coefficient.
Area of Science:
- Materials Science
- Nanotechnology
- Drug Delivery
Background:
- Layered double hydroxides (LDHs) are versatile materials for drug intercalation.
- Controlling the synthesis of drug-loaded nanomaterials is crucial for effective drug delivery.
Purpose of the Study:
- To investigate the synthesis of methotrexatum intercalated layered double hydroxides (MTX/LDHs) using reverse microemulsions.
- To explore the influence of synthesis parameters on MTX/LDHs properties and anticancer efficacy.
- To establish a correlation between drug efficacy and material characteristics.
Main Methods:
- Reverse microemulsion method for MTX/LDHs synthesis.
- Characterization using XRD, FTIR, TEM, and AFM.
- In vitro drug release studies in PBS (pH 7.4).
- Anticancer efficacy evaluation using MTT assay on A549 cells.
Main Results:
- Successful intercalation of methotrexatum (MTX) into LDHs confirmed by XRD and FTIR.
- Synthesis parameters (temperature, water content, MTX content) influence particle structure and size.
- Drug release followed a parabolic diffusion model, indicating a multi-phase ion-exchange process.
- MTX/LDHs hybrids demonstrated anticancer efficacy against A549 lung cancer cells.
Conclusions:
- Reverse microemulsions enable controlled synthesis of MTX/LDHs hybrids.
- Particle properties and drug release kinetics are tunable via synthesis conditions.
- Drug efficacy is significantly correlated with the dispersion coefficient of the MTX/LDHs hybrids, offering new insights for drug delivery system design.

Aldehydes and Ketones with Water: Hydrate Formation
The formation of hydrates is a reversible reaction. Hydrate formation is influenced by steric and electronic factors accompanying the alkyl substituents on the carbonyl group: The rate of hydrate formation increases with a decrease in the number of alkyl groups attached to the carbonyl carbon. Hence,...
