A new high-throughput assay effectively measures superoxide anion radical scavenging capacity. This method is validated for screening antioxidants in natural products and food extracts.
Area of Science:
- Biochemistry
- Analytical Chemistry
Background:
- Superoxide anion radical (O2(•-)) is a reactive oxygen species implicated in oxidative stress.
- Accurate and efficient assays are needed to quantify antioxidant capacity against O2(•-).
Purpose of the Study:
- To develop and validate a high-throughput assay for superoxide anion radical scavenging capacity.
- To optimize reaction conditions for assay reliability and sensitivity.
Main Methods:
- Utilized the xanthine oxidase/xanthine reaction system for O2(•-) generation.
- Optimized parameters including wavelength, reactant concentrations, temperature, time, pH, and solvents.
- Validated assay performance through linearity, precision, accuracy, and stability assessments.
Main Results:
- Achieved high linearity (r(2) = 0.9513-0.9957) and good precision (intraday RSD 1.13-4.05%, interday RSD 2.13-5.62%).
- Demonstrated accuracy (95.64-97.42% recovery) and stability.
- Showed high correlation (r > 0.99) with a conventional colorimetric method.
Conclusions:
- The developed high-throughput assay is accurate, reliable, and efficient for measuring O2(•-) scavenging capacity.
- Suitable for screening large numbers of samples, including food extracts and natural products.
- Facilitates the identification of potential superoxide anion radical scavengers with minimal sample requirements.
