This study presents a safe and cost-effective rat model for spinal fusion research. While mesenchymal stem cells (MSCs) showed minor anti-inflammatory effects, they did not significantly enhance new bone formation in this model.
Area of Science:
- Bone biology and regenerative medicine
- Physiological research focusing on stem cell applications
Background:
- Spinal fusion is a significant challenge in bone biology.
- Osteoinduction and new bone formation are key indicators in ectopic models.
- Mesenchymal stem cells (MSCs) are being investigated for their regenerative potential.
Purpose of the Study:
- To establish a simple, safe, and inexpensive rat model for spinal fusion.
- To evaluate the efficacy of titanium microplates with hydroxyapatite scaffolds combined with human bone marrow-derived MSCs for spinal fusion.
Main Methods:
- 15 Wistar rats underwent interspinous fixation of adjacent vertebrae.
- Group 1: Augmentation material + collagen membrane.
- Group 2: Augmentation material + 5 × 10⁶ MSCs + collagen membrane.
- Histology, histomorphometry, and micro-CT analysis were performed at 8 weeks.
Main Results:
- The interspinous fusion model was safe, inexpensive, and showed zero mortality.
- No significant pathological changes or tumor formation were observed.
- A non-significant difference in new bone formation between groups was noted.
- Minor inflammatory response in the MSC group suggested immunomodulatory effects.
Conclusions:
- The developed rat model for interspinous spinal fusion is practical and easy to implement in small animals.
- While MSCs demonstrated potential anti-inflammatory properties, their impact on bone formation in this specific model was not statistically significant.
