Bone morphogenetic protein 2 (BMP2) induces miR-302a, which targets transcription factor COUP-TFII. This promotes osteoblast differentiation and bone formation by repressing COUP-TFII.
Area of Science:
- Molecular Biology
- Cell Biology
- Biochemistry
Background:
- Chicken ovalbumin upstream promoter transcription factor II (COUP-TFII) inhibits osteoblast differentiation.
- Bone morphogenetic protein 2 (BMP2) suppresses COUP-TFII expression during osteoblast differentiation.
Purpose of the Study:
- Identify BMP2-regulated and COUP-TFII-targeting microRNAs (miRNAs).
- Investigate the regulatory roles of these miRNAs in osteoblast differentiation.
Main Methods:
- In silico analysis to select candidate miRNAs.
- Quantitative real-time PCR to examine miRNA and mRNA expression.
- Luciferase assay to confirm miRNA-target interaction.
- Cell culture and transfection to assess functional effects.
Main Results:
- BMP2 induced miR-302a expression in a dose- and time-dependent manner, decreasing COUP-TFII levels.
- Runx2 transcription factor regulated miR-302a expression.
- miR-302a directly targeted COUP-TFII mRNA at two binding sites.
- Overexpression of miR-302a enhanced osteogenic marker gene expression and activity, while inhibiting it had the opposite effect.
Conclusions:
- miR-302a is induced by osteogenic stimuli and promotes osteoblast differentiation.
- miR-302a acts as a positive regulator of osteoblast differentiation by targeting and repressing COUP-TFII.
