Summary

Cefixime concentrations in human lung tissue were measured to assess its effectiveness against pulmonary infections. Results indicate that a 400 mg daily dose of cefixime may effectively treat lung infections.

Area of Science:

  • Pharmacology
  • Infectious Diseases
  • Respiratory Medicine

Background:

  • Pulmonary infections require effective antibiotic treatment.
  • Understanding antibiotic concentrations in lung tissue is crucial for treatment efficacy.
  • Cefixime is a third-generation cephalosporin with activity against common respiratory pathogens.

Purpose of the Study:

  • To measure cefixime concentrations in human lung tissue in vivo.
  • To evaluate the potential effectiveness of cefixime in treating pulmonary infections.
  • To determine optimal cefixime dosing for lung infections based on pharmacokinetic data.

Main Methods:

  • Twenty-three patients undergoing lung surgery received oral cefixime (200 mg BID or 400 mg QD).
  • Blood and lung tissue samples were collected 4 or 8 hours post-dose.
  • Cefixime concentrations were quantified using High-Performance Liquid Chromatography (HPLC).

Main Results:

  • Significant penetration of cefixime into both normal and neoplastic lung tissue was observed.
  • Lung cefixime concentrations at 4 or 8 hours post-dose exceeded MIC90 values for susceptible bacteria.
  • Pharmacokinetic data supported therapeutic activity at tested dosages.

Conclusions:

  • Cefixime demonstrates significant lung tissue penetration.
  • The measured concentrations suggest cefixime is effective against sensitive pathogens in the lung.
  • A once-daily 400 mg dose of cefixime appears potentially effective for treating pulmonary infections.
