Area of Science:

  • Analytical Chemistry
  • Pharmacology

Background:

  • Accurate quantification of methylprednisolone in human plasma is crucial for pharmacokinetic studies.
  • Existing methods may be laborious or costly.

Purpose of the Study:

  • To develop and validate an economical, reproducible, and automated method for methylprednisolone quantification in human plasma.
  • To apply the developed method to a pharmacokinetic study.

Main Methods:

  • Online solid-phase extraction coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS).
  • Sample pretreatment involved protein precipitation and methanol/water dilution.
  • HySphere C8 EC-SE cartridge for SPE and Agilent Eclipse XDB column for separation.
  • Electrospray ionization in positive ion mode with multiple reaction monitoring (MRM).

Main Results:

  • The method demonstrated excellent selectivity, precision (RSD within 4.45%), and accuracy.
  • Linear calibration curve from 5.25 to 525 ng/mL.
  • The method is less laborious, faster, and has lower consumable costs per sample.
  • Successfully applied to a pharmacokinetic study involving oral administration of methylprednisolone.

Conclusions:

  • The developed online SPE-LC-MS/MS method is a robust, efficient, and cost-effective approach for quantifying methylprednisolone in human plasma.
  • This validated method is suitable for pharmacokinetic analysis and clinical applications.
