Summary

Early detection of implant inflammation may be possible through blood tests. High levels of interferon-gamma (IFNγ) in early blood samples correlated with a greater inflammatory response around titanium implants in rats.

Purpose of the Study:

  • To investigate the association between serum cytokine profiles and peri-implant tissue inflammation.
  • To determine if early serum inflammatory markers can predict late-stage local immune responses to titanium implants.

Main Methods:

  • Rats received intramuscularly implanted, surface-modified titanium (Ti) implants.
  • Serum concentrations of pro-inflammatory (IFNγ, IL-2) and anti-inflammatory (IL-4, IL-10) cytokines were measured post-implantation.
  • Peri-implant tissue response was assessed at 56 days, quantifying CD68+, CD163+, MHC class II+, NK cells, and mast cells.
  • Multivariate and Pearson correlation analyses were employed to assess relationships between serum markers and tissue responses.

Main Results:

  • A significant positive correlation was found between early serum interferon-gamma (IFNγ) levels and the late-stage presence of CD68-positive monocytes/macrophages.
  • This correlation was strongest within the first three weeks post-implantation.
  • No significant associations were observed between other measured cytokines (IL-2, IL-4, IL-10) and the evaluated peri-implant cell types.

Conclusions:

  • Early-stage serum IFNγ levels may serve as a predictive biomarker for the development of pro-inflammatory responses at titanium implant sites.
  • Non-invasive monitoring of serum IFNγ could potentially aid in predicting and managing inflammation-related complications associated with biomaterial implants.
  • Further research in patient samples is warranted to validate these findings for clinical application.
