Position-effect Variegation02:32

Position-effect Variegation

7.3K
In 1928, a German botanist Emil Heitz observed the moss nuclei with a DNA binding dye. He observed that while some chromatin regions decondense and spread out in the interphase nucleus, others do not. He termed them euchromatin and heterochromatin, respectively. He proposed that the heterochromatin regions reflect a functionally inactive state of the genome. It was later confirmed that heterochromatin is transcriptionally repressed, and euchromatin is transcriptionally active chromatin.

Background and Environment Affect Phenotype02:27

Background and Environment Affect Phenotype

8.2K
Although the genetic makeup of an organism plays a major role in determining the phenotype, there are also several environmental factors, such as temperature, oxygen availability, presence of mutagens, that can alter an organism’s phenotype.
An example of how genetic background affects phenotype can be seen in horses. The Extension gene in horses is responsible for their coat color. A wild-type gene (EE) produces black pigment in the coat, while a mutant gene (ee) produces red pigment. A...

Area of Science:

  • Developmental Biology
  • Molecular Biology
  • Genetics

Background:

  • Bone Morphogenetic Proteins (BMPs) are crucial signaling molecules regulating animal development.
  • BMP signaling involves Smad transcription factors and controls gene expression.
  • In Drosophila, Decapentaplegic (Dpp) signaling is modulated by the transcription factor brinker (brk).

Purpose of the Study:

  • To investigate BMP-dependent gene regulation during Drosophila oogenesis.
  • To identify regulatory sequences controlling brinker (brk) and broad (br) gene expression.
  • To elucidate the transcriptional cascade connecting Dpp signaling to target gene regulation in eggshell patterning.

Main Methods:

  • Analysis of cis-regulatory sequences for brk and br.
  • Investigating the roles of Smad and Schnurri (Shn) transcription factors.
  • Studying the epistatic relationship between brk and Dpp in br expression.

Main Results:

  • Dpp directly represses brk transcription via Smad and Schnurri.
  • brk acts epistatically to Dpp in regulating br expression.
  • brk indirectly activates br by removing an unknown repressor.

Conclusions:

  • This study provides novel cis-regulatory insights into BMP signaling interpretation during oogenesis.
  • A transcriptional cascade linking Dpp to target gene regulation in eggshell morphogenesis is defined.
  • The findings enhance understanding of developmental gene regulation and signaling pathways.
