Researchers isolated two new triterpenoid saponins from Ilex cornuta stems. These compounds, along with two known ones, were tested for protective effects against H9c2 cardiomyocyte injury but showed no efficacy.
Area of Science:
- Natural Products Chemistry
- Pharmacology
- Cardiovascular Research
Background:
- Ilex cornuta is a plant species known to contain various bioactive compounds.
- Triterpenoid saponins are a class of natural products with diverse pharmacological properties.
- Cardiomyocyte injury models are crucial for understanding and treating heart diseases.
Purpose of the Study:
- To isolate and structurally elucidate new triterpenoid saponins from Ilex cornuta stems.
- To evaluate the potential cardioprotective effects of isolated compounds against hydrogen peroxide-induced injury in H9c2 cells.
Main Methods:
- Isolation of compounds using chromatographic techniques.
- Structural determination through spectroscopic methods, including 1D/2D NMR and HR-ESI-MS.
- Assessment of cell-protective effects using an in vitro model of H9c2 cardiomyocyte injury induced by H2O2.
Main Results:
- Two novel triterpenoid saponins, identified as ursane-12,19-diene-28-oic acid 3β-O-β-D-glucuronopyranoside-6-O-methyl ester (1) and 3α,23α-dihydroxy-olean-9(11),12-diene-28-oic acid 28-O-β-D-glucopyranosyl ester (2), were isolated.
- Two known triterpenoids (3 and 4) were also identified.
- None of the tested compounds (1-4) demonstrated any significant cell-protective effect against H2O2-induced H9c2 cardiomyocyte injury.
Conclusions:
- The chemical investigation of Ilex cornuta stems led to the identification of two new triterpenoid saponins and two known triterpenoids.
- The isolated triterpenoid saponins and known triterpenoids from Ilex cornuta did not exhibit cardioprotective activity in the tested in vitro model.
