Area of Science:

  • Biomedical research
  • Tissue preservation techniques
  • Neuromuscular disease diagnostics

Background:

  • Muscle biopsy samples require immediate freezing for analysis, often necessitating patient travel to specialized labs.
  • Current preservation methods limit sample transport time, posing challenges for patients with neuromuscular diseases.

Purpose of the Study:

  • To develop and evaluate a novel preservative solution for striated muscle tissue.
  • To assess the protective capabilities of the solution on tissue morphology and molecular integrity over time.

Main Methods:

  • A preservative solution, Kurt-Ozcan (KO) solution, was prepared and tested on rat striated muscle samples.
  • Samples were analyzed for morphological, enzyme histochemical, molecular, and biochemical changes at various time points (3, 6, 18, 24 hours).
  • Statistical analysis included Chi-square and Kruskal Wallis tests.

Main Results:

  • KO solution maintained excellent morphological and enzyme activity scores for up to 18 hours.
  • No significant differences were observed between KO-preserved samples and controls at 18 hours.
  • Messenger RNA (mRNA) integrity was preserved for up to 6 hours in KO solution.

Conclusions:

  • The KO solution effectively preserves the morphological, enzyme histochemical, and biochemical characteristics of striated muscle tissue for 18 hours.
  • The solution also maintains mRNA stability for 6 hours, facilitating further molecular analysis.
  • This offers a viable option for preserving muscle biopsy samples during transport.
