This study identified various forms and modifications of thymosin beta (β) and alpha (α) peptides in tissues and fluids. Further research is needed to understand the clinical significance of these thymosin proteoforms.
Area of Science:
- Biochemistry
- Proteomics
- Molecular Biology
Background:
- Thymosins are a family of peptides involved in various cellular processes.
- Understanding thymosin proteoforms is crucial for their role in health and disease.
Purpose of the Study:
- To characterize β and α thymosins and their proteoforms in diverse tissues and bodily fluids.
- To investigate the association of thymosin proteoforms with various pathologies.
Main Methods:
- Utilized a top-down proteomic platform.
- Employed high-performance liquid chromatography (HPLC) coupled with high-resolution LTQ-Orbitrap mass spectrometry (MS).
Main Results:
- Identified thymosin β4 (Tβ4) and β10 (Tβ10), including post-translational modifications and C-terminal truncated forms.
- Characterized sulfoxide and acetylated derivatives of Tβ4 and Tβ10.
- Detected diverse proteoforms and proteolytic fragments of prothymosin α, parathymosin α, thymosin α1, and thymosin α11.
Conclusions:
- The study identified numerous thymosin proteoforms and modifications across various biological samples.
- The clinical and prognostic significance of these thymosin proteoforms requires further in-depth investigation.
