Regulation of Hematopoietic Stem Cells01:01

Regulation of Hematopoietic Stem Cells

4.4K
All blood and immune cells are produced from the multipotent hematopoietic stem cells (HSCs) by the process of hematopoiesis. However, they all have a limited life span. In addition, many are depleted in immune surveillance or combatting an injury or infection. This makes blood one of the most regenerative tissues. Hematopoiesis helps replenish these blood and immune cells, restoring the body's normal functioning. However, overproduction of blood and immune cells can make them cancerous or...

Mesenchymal Stem Cells01:19

Mesenchymal Stem Cells

6.0K
Mesenchymal stem cells (MSCs) are adult stem cells that can differentiate into most connective tissue cell types, except for hematopoietic cells, depending upon the source of MSCs. For example, bone-marrow-derived MSCs (BM-MSCs) can differentiate into osteocytes, hepatocytes, and pancreatic and neuronal cells. MSCs can be isolated from various sources such as bone marrow, placenta, adipose tissue, teeth, and Wharton’s jelly, a gelatinous substance in the umbilical cord. The ease of their...

Role of Hematopoietic Growth Factors01:28

Role of Hematopoietic Growth Factors

4.4K
Hematopoietic growth factors are molecules that regulate the differentiation rate of hematopoietic stem cells (HSCs). Erythropoietin (EPO), primarily produced by the kidneys, plays a crucial role in erythrocyte production. When oxygen levels in the blood are low, EPO is released into the bloodstream, reaching the bone marrow, where it stimulates HSCs to differentiate and mature into erythrocytes, which are vital for oxygen transport.
Thrombopoietin (TPO), mainly released by the liver,...

Multipotency of Hematopoietic Stem Cells01:19

Multipotency of Hematopoietic Stem Cells

4.1K
The hematopoietic stem cells or HSCs are multipotent, meaning they can differentiate and give rise to all blood and immune cells. HSCs are maintained in the quiescent stage until an external stimulus initiates their differentiation. The multipotent HSCs exist as two heterogeneous populations, long-term repopulating cells (LTRC) and short-term repopulating cells (STRC). The two HSC populations have different surface markers or receptors and are classified based on quiescence and long-term...

Clinical Applications of Epidermal Stem Cells01:19

Clinical Applications of Epidermal Stem Cells

3.4K
Epidermal stem cells (EpiSCs) are mainly located at the basal layer of the epidermis. These cells repair minor injuries of the skin and replace dead skin cells. However, EpiSCs’ cannot heal severe wounds such as major burns or those from diabetes or hereditary disorders. In such cases, culturing the epidermal stem cells from the patient is possible and has yielded successful treatment options, such as laboratory-grown skin grafts. These grafts are synthesized using a patient’s own...

Area of Science:

  • Immunology
  • Neuroscience
  • Psychiatry

Background:

  • Hematopoietic stem cell transplantation (HCT) presents significant emotional and immune challenges.
  • The endocannabinoid (eCB) signaling system modulates immune responses and emotional reactivity.
  • The role of the eCB system during HCT remains largely uncharacterized.

Purpose of the Study:

  • To investigate the function of the eCB signaling system in HCT recipients.
  • To assess changes in eCBs, inflammatory molecules, and psychosocial factors peri-transplant.
  • To explore associations between these variables during the HCT recovery process.

Main Methods:

  • 19 HCT recipients were studied at three peri-transplant time points.
  • Psychosocial data and blood samples were collected.
  • Circulating eCBs, inflammatory markers (IL-6, CRP), and psychosocial factors were analyzed.

Main Results:

  • HCT recipients showed significant changes in inflammation and psychosocial functioning, but not circulating eCB levels.
  • Associations between psychosocial factors and inflammation were strongest pre-transplant.
  • The eCB 2-arachidonoylglycerol (2-AG) correlated positively with IL-6 and CRP, and negatively with depressive symptoms.

Conclusions:

  • The eCB system's regulation during HCT may involve mechanisms beyond the immune system.
  • Significant associations between eCBs, inflammation, and depressive symptoms warrant further investigation.
  • Understanding the eCB system is crucial for managing complex procedures like HCT.
