A new modulated explant/enzyme method (MEEM) efficiently isolates millions of high-quality mesenchymal stem cells (MSCs) from umbilical cords. This cost-effective technique yields sufficient MSCs for regenerative medicine applications within a month.
Area of Science:
- Stem Cell Biology
- Regenerative Medicine
- Biotechnology
Background:
- Mesenchymal stem cells (MSCs) are crucial for regenerative medicine.
- The umbilical cord (UC) is a promising source of MSCs due to its abundance and uniformity.
- Standardized, high-yield MSC isolation methods are needed to meet increasing demand.
Purpose of the Study:
- To develop and present a modulated explant/enzyme method (MEEM) for maximizing MSC isolation from the entire UC.
- To establish a reproducible and cost-efficient protocol for large-scale MSC production.
- To characterize the isolated MSCs for their potential in cell therapy.
Main Methods:
- A novel modulated explant/enzyme method (MEEM) was employed to isolate MSCs from all regions of the UC.
- Primary culture was performed over 8–10 days, followed by expansion for 28–30 days through three passages.
- Characterization included immunophenotyping (CD markers) and gene expression analysis (SOX2, OCT4, NANOG), alongside differentiation assays.
Main Results:
- MEEM successfully isolated 6–10 million MSCs in the primary culture within 8–10 days.
- After three passages, 8–10 × 10^8 cells were obtained in 28–30 days.
- Isolated MSCs expressed key markers (CD73, CD90, CD105, CD44, SOX2, OCT4, NANOG) and differentiated into adipocytes and osteocytes.
Conclusions:
- The MEEM is a simple, reproducible, and cost-efficient method for isolating large quantities of high-quality MSCs from UC.
- This method enables the production of sufficient MSCs for cell therapy within a month.
- The characterized MSCs hold significant potential for applications in regenerative medicine.
