Hydroxychloroquine (HCQ) reduces microglial activation and pro-inflammatory cytokine production in vitro. In vivo, HCQ delayed experimental autoimmune encephalitis onset and reduced demyelination, suggesting potential for multiple sclerosis treatment.
Area of Science:
- Neuroimmunology
- Pharmacology
Background:
- Microglial activation is central to multiple sclerosis (MS) pathogenesis.
- Hydroxychloroquine (HCQ), an antimalarial drug, possesses immunomodulatory effects.
- This study investigates HCQ's impact on microglial activation and experimental autoimmune encephalitis (EAE).
Purpose of the Study:
- To evaluate the effect of HCQ on human microglial activation in vitro.
- To assess HCQ's efficacy in preventing or delaying the development of EAE in vivo.
Main Methods:
- Human microglia were activated with lipopolysaccharide (LPS).
- Cytokine production in HCQ-treated and untreated microglia was measured.
- HCQ's effect on EAE development and spinal cord pathology was assessed in mice.
Main Results:
- HCQ significantly reduced pro-inflammatory (TNF-α, IL-6, IL-12) and anti-inflammatory (IL-10, IL-1 receptor antagonist) cytokine production by LPS-stimulated microglia.
- HCQ pretreatment delayed EAE onset in mice.
- Reduced microglial/macrophage infiltration and demyelination were observed in HCQ-treated animals' spinal cords.
Conclusions:
- HCQ effectively suppresses human microglial activation in vitro.
- HCQ demonstrates neuroprotective effects by delaying EAE and reducing spinal cord pathology.
- HCQ is a promising candidate for clinical investigation in multiple sclerosis therapy.
